Die statistischen Probleme der Evaluation von in vitro-Verfahren zur Klassifikation von Substanzen entsprechend ihrer in vivo-Toxizität werden geschildert und mit den Ergebnissen eines Ringversuchs beispielhaft demonstriert, bei dem die Möglichkeit eines Ersatzes des Draize-Tests durch den HET-CAM-Test und einen Zytotoxizitätstest (3T3-Zellen, Neutralrot-Assay (NRU)) untersucht wurde. Insbesondere werden die statistischen Verfahren der Baumanalyse (CART) zur Aufstellung optimaler Klassifikationsregeln erörtert.

Biometrical methods for the evaluation of in vitro-tests
The statistical problems of the evaluation of in vitro tests for the classification of compounds according to their in vivo toxicity are discussed. The main problem is the establishment of classification rules (based on the outcomes of the in vitro test) with highest probability for correct classification. The classical statistical classification methods, the linear or nonlinear discriminant analysis, suffer from inflexibility to handle combinations of in vitro endpoints. The disadvantage is overcome with the technique of ‘Classification and Regression Trees’ (CART). This method is demonstrated and compared with the technique of discrimmant analysis with the results of a ring-experiment for the evaluation of the HET-CAM-test and NRU-test (with 3T3-cells) as replacement of the Draize-test with rabbit-eyes.
