Zusammenfassung
Aviäre vitelline Antikörper können nach Immunisierung von Hühnern mit Proteinantigen induziert und unkompliziert, unblutig aus dem Eidotter gewonnen werden.
In immunologischen Testsystemen werden üblicherweise Sekundärreagenzien eingesetzt, da Primärantikörper meist nicht direkt markiert (zum Nachweis der Antigen-Antikörper-Reaktion) vorliegen und indirekte Nachweissysteme (Antigen-Primärantikörper-markierter Sekundärantikörper) den Vorteil einer höheren Empfindlichkeit bieten.
Mit der Herstellung von Huhn-Antikörpern (IgY) gegen Mausimmunglobuline (unterschiedliche Isotypen und Subklassen) konnte gezeigt werden, daß nach geeignetem Immunisierungsmodus qualitativ hochwertige Antikörperpräparate zum Erfassen von monoklonalen Mausantikörpern zur Verfugung stehen.
Als Einsatzmöglichkeiten für die Huhn-anti-Mausimmunglobulin-Konjugate (markiert mit Fluoreszenzfarbstoff, Biotin bzw. Enzymen) werden Varianten der Durchflußzytometrie und bei ELISA-Systemen vorgestellt. Mit dem Nachweis der Eignung aviärer vitelliner Antikörper als Sekundärreagens wird eine Alternative zur Verwendung mammärer Antikörper, die auf blutige Art und Weise gewonnen werden müssen, aufgezeigt.
Summary
After immunization of hens with protein antigen avian vitelline antibodies are induced. It is uncomplicated to get these antibodies from egg yolk by an unbloody way.
Secondary reagents are commonly used in immunological tests because primary antibodies often not directly conjugated and indirect systems are more sensitive in detecting antigen antibody reactions.
It is shown that after immunisation of hens with mouse immunoglobulins (different isotypes and subclasses) IgY could be prepared in great amounts. The IgY preparates are excellent tools in detecting monoclonal mice antibodies.
Conjugates of IgY (fluorescence dye, biotin or enzymes) are tested in FACS analysis and ELISA. It is demonstrated that avian vitelline secondary antibodies are a good alternative of mammalian secondary antibodies.
