Analytical reliability of methods to determine theophylline in serum or plasma

Abstract

Theophylline is frequently used in patients with acute or chronic reversible airway disease. Its analysis in serum or plasma is required both for pharmacological studies and the individual control of drug therapy. Therapeutic drug monitoring is necessary because of wide individual variation of the liver capacity to metabolize the drug under various physiologic or pathologic circumstances. Five basic methods are recommended for the analysis of theophylline in serum or plasma, most of them in many variants:

  1. 1.

    Extraction and UV-spectrophotometry (UV) [6, 18, 19]

  2. 2.

    Radioimmunoassay (RIA) [2, 11)

  3. 3.

    High pressure liquid chromatography (HPLC) [3, 10, 13, 14, 22, 23, 24]

  4. 4.

    Homogeneous enzymeimmunoassay (EIA, EMIT®) [4, 5, 8, 12]

  5. 5.

    Gaschromatography (GC) [7, 9, 15, 16, 20, 21]

We evaluated the methods 1–4 with regard to analytical reliability, reagent cost and practicability.
