Zusammenfassung
Die systembiologische Modellierung von Signalkaskaden und Protein-Netzwerken erlaubt vertiefte Einblicke in die Funktion der beteiligten Proteine und hilft dadurch Krankheitsursachen zu verstehen, Infektionsprozesse und Immunantworten besser zu beschreiben oder auch komplexe Vorgänge in der Biologie, wie Zelldifferenzierung und Neurobiologie, aufzuklären. Stärker mathematische Modelle beschreiben Signalnetzwerke genau in der Änderung im Zeitverlauf und ihre Geschwindigkeit mithilfe von Differentialgleichungen. Dies erklärt den Vorgang genau, verbringt aber zusätzlich Zeit z. B. mit dem Ermitteln der Geschwindigkeiten (Kinetik; Zeitreihenanalyse). Boole’sche Modelle benötigen dagegen als Information nur welche Proteine im Netzwerk beteiligt sind und welches Protein mit welchen anderen Proteinen wie interagiert (aktivierend oder hemmend). Auf einem Booleschen Modell aufbauende Simulationen (z. B. mit SQUAD oder Jimena) müssen iterativ in vielen Zyklen überprüft werden, ob denn das Verhalten in dem Computermodell auch zu dem tatsächlich im Experiment beobachteten Ausgang zumindest qualitativ passt. Das Computermodell wird also Schritt für Schritt an die Daten angepasst (Data-Driven-Modeling; Daten-getriebene Modellierung).
