Zusammenfassung
Das Verständnis pathophysiologischer Grundlagen der Entstehung von epileptischen Anfällen und Epilepsien ist Voraussetzung für die Entwicklung neuer Therapiekonzepte. Aus ethischen Gründen sind der Forschung an Menschen enge Grenzen gesetzt, daher muss häufig auf Tiermodelle zurückgegriffen werden. Die Entstehung eines akut symptomatischen Anfalls ist – in Abhängigkeit von der Ursache – in der Regel auf eine reversible Verschiebung im Verhältnis von Inhibition zu Exzitation zurückzuführen. Bei Epilepsien stehen eher die Mechanismen, die zur Epileptogenese beigetragen haben, im Vordergrund. Diese führen über ätiologieabhängige Wege zu einer reduzierten Anfallsschwelle, die das Auftreten von unprovozierten Anfällen erleichtert. Tiermodelle sollen zur Erforschung pathophysiologischer Mechanismen beitragen, sie dienen zudem zum Screening von neuen potenziell antiepileptisch wirksamen Substanzen. Kein Modell ist ideal, es werden je nach Fragestellung unterschiedliche Modelle eingesetzt.
