Abstract
Among other biophysical techniques, attenuated total reflection–Fourier transform infrared (ATR-FTIR) spectroscopy has widely been employed to characterise the skin barrier. Major advantages are its non-destructive and time-saving nature. Spectra of the stratum corneum depict characteristic bands such as the CH2-stretching and CH2-scissoring as well as the amide 1 and 2 vibrations. While the latter describe the secondary structure of the stratum corneum proteins and may reflect upon the water absorption, the CH2-stretching and scissoring bands give information about the lateral packing of the skin lipids. This chapter provides an introduction on the characterisation of the stratum corneum by ATR-FTIR spectroscopy and an overview of the influence of selected permeation enhancers and drug delivery systems on the skin barrier as investigated by this technique.
