Summary
In adult rats single high doses of ENBU administered by stomach tube induced squamous carcinomas of the forestomach and in two of 22 animals adenocarcinomas of the glandular stomach. In transplacental experiments one single dose of 100 mg or 200 mg/kg was given to pregnant rats on the 15th or 22nd day of gestation respectively. With the exception of four all other offspring (118) later died with malignant tumours of the nervous system. When given to rats ten days old as a single subcutaneous injection of 40 or 80 mg/kg neurogenic malignomas also developed in all rats treated. The distribution of the tumours within the various parts of the nervous system, however, showed characteristic differences dependent on the time of ENBU administration during prenatal and neonatal development. The importance of the time factor in carcinogenesis is briefly discussed.
Zusammenfassung
Bei erwachsenen Ratten erzeugte ÄNBU nach einmaliger Gabe hoher Dosen mit Schlundsonde Plattenepithelkrebs des Vormagens und in 2 von 22 Fällen Adenocarcinome des Drüsenmagens. In transplacentalen Versuchen erhielten schwangere Ratten am 15. bzw. 22. (letzten) Tage der Gravidität einmalig 100 bzw. 200 mg/kg. Mit Ausnahme von 4 Tieren starben alle anderen 118 Nachkommen später mit malignen Tumoren des Nervensystems. Nach einmaliger subcutaner Injektion von 40 bzw. 80 mg/kg an 10 Tage alte Ratten wurden ebenfalls neurogene Malignome bei allen behandelten Tieren beobachtet. Die Verteilung der Tumoren auf die einzelnen Teile des Nervensystems zeigte indessen charakteristische Unterschiede je nach dem Zeitpunkt der Gabe von ÄNBU während der vorgeburtlichen oder postnatalen Entwicklung. Die Bedeutung des Zeitfaktors bei der Krebserzeugung wird kurz diskutiert.
