Abstract

Leukocyte adhesion to vascular endothelium is a hallmark of the inflammatory process. In recent years, many studies have been designed to address the cellular and molecular mechanisms involved with mediating the adhesive interactions between leukocytes and vascular endothelial cells. Significant progress in our understanding of these adhesion mechanisms has resulted from technological advances in our ability to grow endothelial cells as monolayers and the production of monoclonal antibodies that bind to and inhibit the function of adhesion molecules on the surface of leukocytes and endothelial cells. While the information gained from studying the expression of adhesion molecules in vitro has improved our understanding of the mechanisms that mediate the expression of cell adhesion molecules in inflamed tissue, there is a concern that monolayers of cultured endothelial cell may not adequately simulate the in vivo situation.
