Zusammenfassung
Die Thrombozyten (Blutplättchen) lösen sich im Knochenmark durch Fragmentation aus dem Plasmaverband der Megakaryozyten heraus. Die Ausgangszelle der Megakaryopoese differenziert sich aus der gemeinsamen hämatopoetischen Stammzelle und wird als Megakaryoblast bezeichnet. Dieser reift unter stetiger Vergrößerung seines Zytoplasmavolumens durch einen endomitotischen Prozeß mit mehreren synchronen Kernverdopplungsschritten zum Megakaryozyten heran. In einem unterschiedlichen Stadium dieses Entwicklungsvorgangs, in der Regel nach Ausbildung von acht Zellkernen, kommen Kernvermehrung und Wachstum des Megakaryozyten zum Stillstand. Sein Zytoplasma nimmt ein granuläres Aussehen an, woraufhin es zur Herauslösung der Thrombozyten kommt (Abb. 11.1). Dieser Vorgang wird durch die Bildung von Mikrovesikeln im Zytoplasma vorbereitet, die miteinander verschmelzen und sich dadurch zu sogenannten Plättchendemarkations-Membranen entwickeln. Aus jedem Megakaryozyten entstehen auf diese Weise ca. 4000 Thrombozyten. Der Zeitraum zwischen Differenzierung der Stammzelle und Thrombozytenbildung beträgt beim Menschen ca. 10 Tage.
