Immunofluorescence: Basic Considerations

Immunofluorescence involves the staining of cells with antibodies and other specific ligands directly or indirectly labeled with fluorescent dyes. This offers fascinating possibilities for the characterization of cells according to the expression of protein or polysaccharide markers specific for differentiation or activation stages. A few basic considerations will help one to understand the important parameters and to optimize the specificity and sensitivity of immunofluorescence staining. Of the three steps in immunofluorescence analysis-staining, measurement, and data evaluation-the first, preparation of cells and staining, is probably most important and therefore receives the most attention.
