The use of epoxy resins as embedding media for electron microscopy

During a study of fixation of bacteria for sectioning and electron microscopy using the conventional methacrylate embedding technique (1) a considerable number of blocks showing almost a 100% burst and swollen cells were noted. By carefully controlling each step in the procedure by means of light microscopy it could be demonstrated that the swelling and bursting occurred during the actual hardening of the methacrylate (2).
