Zusammenfassung
Glukokortikoide werden seit 1948 als antiphlogistische, antiallergische und immunsuppressive Substanzen angewandt. Grundgerüst der Glukokortikoide ist ein polyzyklischer Kohlenwasserstoff, das Pregnan. Die Biosynthese beginnt mit der Oxidation von Cholesterol über Pregnolo-non zu Progesteron. Durch Hydroxylierung an C17 und nachfolgende Hydroxylierung an C21 und C11 entsteht das körpereigene Glukokortikoid, das Kortisol. Wichtig für die biologische Wirkung sind die Doppelbindung zwischen C4 und C5, die Ketogruppe an C3, die zweigliedrige Kohlenstoffseitenkette an C17, die Hydroxyl- bzw. Ketogruppe an C11 und die Hydroxylgruppe an C17 in α-Position.
