Summary
A marginal siderosis of the central nervous system was produced experimentally in rabbits by repeated injections of an iron-dextran solution (Myofer) into the cisterna magna. By light microscopy, an excessive storage of iron could be observed mainly within the leptomeninges and within cells of the superficial cerebral and cerebellar cortex. At the submicroscopic level, the iron deposits in the brain tissue consisted of very small and dense particles which often exhibited the typical sub unit structure of ferritin. These iron-containing granules were found both to lie tightly packed within siderosomes and to be randomly dispersed throughout the ground cytoplasm of the storing cells. It should be emphasized that the deposition of iron took place nearly exclusively in astrocytes and in reactively proliferated microglia cells. Within the cell bodies and cytoplasmic processes of neurons neither siderosomes nor diffusely scattered ferritin granules could hitherto be detected. Particles of high density were never seen within pinocytotic vesicles of subpial astrocytes and of ependymal cells or within the intercellular gaps of the neuropil. It is, therefore, suggested that the iron had entered the brain tissue from the cerebrospinal fluid in an ionic (i. e. soluble) form.
Zusammenfassung
Bei Kaninchen wurde durch wiederholte intrazisternale Injektionen einer Eisen-Dextran-Lösung (Myofer) eine Randzonensiderose des Zentralnervensystems erzeugt. Lichtmikroskopisch zeigte sich eine massive Speicherung von Eisen vor allem in der Leptomeninx sowie in Zellelementen der oberen Groß- und Kleinhirnrindenschichten. Mit dem Elektronenmikroskop konnten im zentralnervösen Gewebe winzig kleine, ungemein kontrastreiche Partikel beobachtet werden, welche vielfach die feineren Struktureigentümlichkeiten des Ferritins aufwiesen. Diese eisenhaltigen Granula fanden sich sowohl dicht gepackt innerhalb von Siderosomen als auch locker verstreut im Grundcytoplasma der speichernden Zellen vor. Hervorzuheben ist, daß die Ablagerung des Metalls weitgehend auf die Astrocyten und auf die lebhaft proliferierende Mikroglia beschränkt blieb. In den Perikaryen und Cytoplasmafortsätzen von Nervenzellen wurden bisher niemals Siderosomen oder einzeln liegende Ferritinkörnchen angetroffen. Micellare Metallteilchen waren weder in Pinocytosevesikeln von subpialen Astrocyten und Ependymzellen noch in den Intercellularfugen des Neuropils festzustellen. Das Eisen dürfte demnach in ionisierter Form aus dem Liquor in das zentralnervöse Gewebe übergetreten sein.
