Zusammenfassung

Von einer nicht zu dichten Flüssigkeitskultur von Pseudomonas (bei Bedarf Verdünnung mit m/15 Phosphatpuffer pH 7,0) wird ein Tropfen auf einen Objektträger gebracht und das Deckglas so aufgelegt, daß in der Flüssigkeit einige kleinere Luftblasen mit eingeschlossen sind. Zur Vermeidung von unerwünschten Strömungen und zum Sauerstoffabschluß umrandet man das Deckglas luftdicht mit geschmolzenem Paraffin. Unter dem Mikroskop wird nun bei mittlerer Vergrößerung (300- bis 500fach) das Verhalten der Bakterien 10–30 min lang beobachtet.
