Zusammenfassung
Die Antikörperbildung erfolgt in den Plasmazellen, die aus den B-Lymphozyten durch Differenzierungs- und Proliferationsvorgänge entstehen. Eine jede Plasmazelle kann nur einen Antikörper gegen ein definiertes Antigen, also nur gegen ein Membran- oder gegen ein lösliches Antigen produzieren. In der immunologischen Abwehr bildet immer ein ganzer Stamm (KLON) von Plasmazellen einen identischen Antikörper, wobei die Syntheseleistung einer einzelnen Plasmazelle bei etwa 2000 Molekülen pro Sekunde liegt. Die biologische Lebensspanne einer einzelnen Plasmazelle begrenzt zeitlich ihre Antikörperproduktion. Bei der Herstellung monoklonaler Antikörper in vitro (18) werden Antikörperproduzierende B-Lymphozyten mit potentiell unsterblichen Myelomzellen der Maus fusioniert. Die weitere Zucht der einzelnen Klone erfolgt in der Kultur oder effektiver in der Aszitesflüssigkeit der Maus.
