Zusammenfassung
Die Transparenz der Cornea ermöglicht besonders gute Beobachtung von Reaktionsabläufen in diesem Gewebe. Daher wurden vergleichende Untersuchungen der Reaktionen intracorneal und in bekannter Weise intracutan injizierter allogener Zellen durchgeführt. Mesenteriallymphknotenzellen nicht sensibilisierter oder durch i.p.-Injektion von LEW-Augen- bzw. -Nierenhomogenaten sensibilisierter CAP-Inzuchtratten (RtH-lc) wurden LEW-Ratten (RtH-11) injiziert, und zwar demselben Empfänger 3×104 intracorneal, sowie 3 × 104, 3 × 105, 3 × 106 und 10×106 intracutan. Die Cutanreaktionen zeigten bei 44 Empfängertieren keine Unterschiede zwischen Zellen vorbehandelter oder normaler allogener CAP-Spender oder syngener LEW-Spender. Demgegenüber zeigten die Cornealreaktionen bei 27 Empfängern ausgeprägtere und länger anhaltende Effekte nach Injektion von Zellen vorbehandelter Spender als nach Applikation normaler allo- oder syngener Zellen. Diese Ergebnisse bedeuten, daß 1:300 der Zellmengc, die intracutan effektlos blieb, gut erkennbare Veränderungen in der Cornea induzierte, d. h. die intracorneale Injektion in der beschriebenen Anordnung eine sehr empfindliche Methode darstellt.
Summary
The fact that the transparency of cornea allows excellent observation of reactions taking place in this tissue led to comparative studies of allogeneic cell transfer reactions in skin and cornea.
Mesenterial lymph node cells of CAP inbred rats (RtH-1c), unsensitized or sensitized by i.p. injections of LEW eye or kidney homogenates, were injected into LEW inbred rats (RtH-11): 3 x 104 intracorneally and 3 x 104, 3 x 105, 3 x 106, 10 x 106 intracutaneously into the same recipients. In 44 recipients the cutaneous reactions of lymph node cells from pretreated donors did not differ significantly from those of normal allogeneic CAP or syngeneic LEW cells. However, in 27 recipients the corneal reactions of lymph node cells from both types of pretreated donors were much more pronounced and longer lasting than the reactions following normal allo- or syngeneic cell injections. These results imply that 1:300 of a cell number which remained without reactions in the skin induced relatively easily visible effects in the cornea and, thus, are discussed as expression of the high sensitivity of the procedure described.
Résumé
Le fait que la transparence de la cornée permet d’observer parfaitement les réactions dont ses tissus sont le siège, a conduit à une étude comparative des réactions de transfer des cellules allogènes dans la peau et la cornee.
Les cellules à nodosité lymphocytaire “Mesenterial” des rats consanguines CAP (RtH-1c) insensibilisés ou sensibilisés par des injections intrapéritoneales de LEW yeux ou reins solutions syngènes, ont été inoculées à des rats consanguins LEW (RtH-11) à 3 x 104 par injection intracornéenne et a 3 x 104, 3 x 105, 3 x 106, 10 x 106 par injections intracutanéennes à un même sujet. Sous 44 sujets les réactions cutanées des cellules à nodosité lymphocytaire des donneurs préalablement traités ne différaient pas de façon significative de celles des cellules allogè nes normales CAP ou homogènes LEW. Cependant chez 27 sujets les réactions cornéennes des cellules à nodosité lymphocytaire des deux types de donneurs prétraités étaient beaucoup plus prononcées et beaucoup plus durables que les réactions normalement provoquée par des injections de cellules allogènes ou syngènes. Ces résultats impliquent que 1/300 du nombre des cellules qui sont demeurées sans réactions dans la peau ont provoqués des effets relativement facilement visibles dans la cornée et pour cette raison, peuvent être considérés comme l’expressions de la haute sensibilité de la procédure dérite.
