Abstract

Cryo-electron microscopy was used to study the appearance of vesicles of phosphatidylcholine. From the micrographs we concluded that lateral tension was produced during the freezing process. Membrane undulations were flattened and also the tension forced the vesicles to change their shapes. However, proper preparation techniques led to the finding of vesicles with grainy membrane surfaces or with pronounced edges or bulges. This supported the idea that unstressed membranes may tend to develop an optically unresolvable superstructure capable of consuming large amounts of membrane area.
