Abstract
The evolutionary origins of DRB pseudogenes have been examined using molecular evolutionary techniques. Data for several primate DRB genes, both functional and pseudogenes were used. Careful consideration was given in determining suitable analytical methods to avoid two confounding effects, namely positive selection for variation within functional loci and the loss of functional constraint in pseudogenes, while allowing the use of data from both coding and non-coding regions. A core data set of exon sequences was used to determine the position of the pseudogenes within the major DRB phylogenetic groupings. The branching order within the pseudogenes themselves was then inferred using supplementary data from introns. The ages of the DRB pseudogenes were estimated using 2 methods which date the origin of the lineage giving rise to DRB4-like pseudogenes at 20–30 mya. Minimum age estimates show that the DRB8 and DRB7 lineages branched from that of DRB2 at least 18–20 mya, followed by DRB6 at about 8–12 mya and DRB4 at about 8 mya.
