Abstract
The liver is a transporting epithelium, composed of sheets of hepatocytes that separate the canalicular lumen from the perisinusoidal space. Transport of macromolecules from blood to bile is thought to occur by a transcellular vesicular pathway (Brown and Kloppel, 1989; Lake et al. 1985), where hepatocytes internalize extracellular material either by specific receptors or by fluidphase endocytosis. Ligands taken up by receptor-mediated endocytosis are selectively delivered to lysosomes (e.g. asialoglycoproteins) or to bile (e.g. transcytosis of polymeric immunoglobulin A). Transcellular vesicular transport of the majority of other blood derived biliary proteins (Reuben, 1984) does not involve specific receptor binding.
