Zusammenfassung
Die biologische Bedeutung der PräS-kodierten Hepatitis B Virus (HBV) — Hüllproteine — insbesondere ihre Regulation innerhalb der Leberzelle — ist noch unklar. Daher war es Ziel unserer Untersuchungen, die PräS- und S-kodierten Hüllproteine vergleichend im Serum und im Lebergewebe auf molekularer Ebene zu untersuchen.
Von asymptomatischen HBsAg-Trägern (n = 20) und Patienten mit chronisch-aktiver Hepatitis B (n = 10) wurden Serum und Leberhomogenat mittels SDS-Polyacryla-midgelelektrophorese aufgetrennt, auf Nitrozellulosemembranen geblottet und anschließend mit monoklonalen Antikörpern gegen PräS1, PräS2 und HBsAg (p24) immungefärbt. Als Ergebnis konnten bei alien Patienten mit chronisch-aktiver Hepatitis B mit Virusreplikation PräS1-, PräS2- und S-kodierte Hüllproteine im Serum nach-gewiesen werden, während im Leberhomogenat hohe Konzentrationen an PräS1- und S-Antigen, aber nur geringe Mengen an PräS2 Antigen zu finden waren. Bei Patienten ohne Virusreplikation überwog im Serum das S-Antigen (p24), während im Lebergewebe vorwiegend PräS1-Protein vorhanden war. Bei asymptomatischen HBsAg-Trägern fanden sich im Serum mit Immunoblot keine PräS-Proteine, sondern nur gering S-Antigen (p24), während im Lebergewebe deutlich das PräSl-Protein erkennbar war. Die molekulare Analyse zeigte zudem, daß das Leber-PräS1 ein niedrigeres Molekular-gewicht als das Serum-PräS1 aufwies (p35/38 gegenüber p39/42). Die PräS- und S-kodierten Hüllproteine sind somit im Serum und in der Leber bei chronischer Hepatitis B unterschiedlich verteilt. Während das PräS1-Protein als Hauptspeicherprotein in der Leber anzusehen ist, stellt das S-Antigen (p24/27) das Hauptsekretionsprodukt im Serum dar. Leber- und Serum-PräSl scheinen zudem biochemisch unterschiedlich zu sein. Die Überexpression des PräS1-Proteins wird auch bei transfizierten Zellinien und im transgenen Mausmodell beschrieben.
