Abstract

PGE2 (5 µg/ml) mimicks the action of corticotrophin upon the rat decapsulated adrenal cortex in vitro, stimulating an increased output of corticosterone (Flack, Jessup and Ramwell, 1969). The effects of submaximal doses of PGE2 and corticotrophin are additive and both are similarly reduced by cycloheximide, an inhibitor of protein synthesis. Cyclic AMP and its dibutyryl analogue also stimulate steroidogenesis and it has been suggested that both PGE2 and corticotrophin exert their effects via a common pathway which includes the adenyl cyclase system (see Chapter XV). Flack and Ramwell (1970) observed a decrease in adrenal prostaglandin concentrations following corticotrophin administration (Fig. 1). This suggests a further interaction between the two substances on steroidogenesis.
