Origin of Peptides Presented by MHC Class I Molecules: Generation by Proteasomes Precedes Evolution of the Immune System
Major histocompatibility complex (MHC) class I molecules present peptide epitopes, derived from cytosolic and nuclear proteins, for recognition by CD8+ T cells. The vast majority of these epitopes are derived from self-proteins and are recognized primarily by the precursors of CD8+ T cells within the thymus, in order to delete those that are self-reactive. A small proportion of MHC class I epitopes, derived from proteins of intracellular pathogens or from mutated tumor-associated self-proteins, are recognized by peripheral CD8 + T cells, which are then activated to cytotoxic cells and destroy the cells presenting these epitopes.
