Zusammenfassung
Mutationen im p53 Tumor-Suppressor Gen, lokalisiert auf Chromosom 17p, sind die häufigsten genetischen Alterationen bei zahlreichen Malignomen des Menschen. Veränderungen im p53-Gen können zu einem Verlust der Wachstumskontrolle durch das p5 3-Protein fuhren, die Zelle proliferiert unkontrolliert und der programmierte Zelltod ist außer Kraft gesetzt. Dadurch kommt es zur Überexpression und Akkumulation von p5 3-Protein in der Zelle, welches als Tumorantigen fungiert und zur Bildung von Antikörpern führen kann. In mehreren Studien wurde gezeigt, daß bei vielen Tumoren eine positive Korrelation der p53 Expression mit der Tumorprognose besteht. Wird im Tumor eine p53 Überexpression oder p53 Mutation nachgewiesen handelt es sich in der Regel um einen aggressiven, wenig oder undifferenzierten und proliferativen Karzinomtyp. Bei bis zu 35% der Tumorpatienten sind Autoantikörper gegen mutiertes p53 nachweisbar, wobei der Prozentsatz abhängig vom Tumortyp ist. Das Ziel unserer Studie war es zu prüfen, ob bei Patienten mit einem Schilddrüsenkarzinom eine Korrelation zwischen dem Tumorstadium und der p5 3-Expression besteht.
