Abstract
The research development of microbial and molecular genetics is followed by the results on the genetics of Rhizobium meliloti and its phages. Two temperate and one virulent Rhizobium phage was characterized. By the established, original phage genetic system genetic and physical maps were constructed, genes identified as well as molecular mechanisms in temperate regulation, phage-bacterium interactions clarified. Transduction was worked out for mapping and for transposon suicide transfer into defective lysogenic recipients. Transformation and transfection was carried out with R. meliloti. The bacterial genome was mapped by transduction and conjugation. The nod and nif genes were localized, characterized, the genetic communication between Rhizobium meliloti and alfalfa revealed with molecular genetic techniques. Plant host specific chemotaxis of Rhizobia was demonstrated and genes involved identified. For practical application and strain improvement a competition test was elaborated including the identification of reisolated symbionts by phage superinfection.
