Zusammenfassung
Die altersabhängige, organspezifische Kinetik von Zeilproliferation und Zellverlust wird bestimmt durch die Proliferationskompetenz der individuellen Zelle im Wechselspiel mit ihrer differenzierten Funktion, durch interzelluläre Kommunikation, Einflüsse der extrazellulären Matrix und der Gewebsstruktur, durch das humorale Mikromilieu unter Mitwirkung auto- und parakriner Faktoren und schließlich durch übergeordnete systemische humorale, hormonelle und zelluläre Faktoren. Gegen dieses hierarchisch geordnete, nur durch kontinuierliche Interaktion zwischen extrazellulären Faktoren und deren zellulären Rezeptoren funktionierende balanzierte System sind Krebszellen auf mindestens einer Ebene refraktär. Sie gewinnen mit der malignen Transformation eine relative, in der Regel fortschreitende Autonomie gegen homöostatische Einflüsse des Organismus bei gleichzeitigem Diffenzierungsverlust.
