The Biologic Significance of Glycoprotein Hormone Oligosaccharides

Abstract

The glycoprotein hormones lutropin (LH), follitropin (FSH), thyrotropin (TSH), and chorionic gonadotropin (CG) are dimeric proteins. They consist of a common a subunit and highly homologous, but hormone-specific, β subunits (Fig. 1). Even though the structures of the glycoprotein hormones are closely related at the primary, secondary, and tertiary levels, the structures of their Asn-linked oligosaccharides differ. LH and TSH bear oligosaccharides which terminate with the unique sequence SO44GalNAcβ1, 4GlcNAcβ1, 2Manα (S4GGnM) whereas FSH and CG bear oligosaccharides which terminate with the sequence sialic acid α2, 3/6Galβ1, 4GlcNAcβ1, 2Manα (Baenziger and Green 1991; Green and Baenziger 1988a, b; Green et al. 1985b; Endo et al. 1979; Mizuochi and Kobata 1980). The synthesis of these sulfated oligosaccharides reflects the activity of two highly specific enzymes, a GalNAc-transferase and a sulfotransferase.
