Molecular Analysis of a Gene (irpA) Encoding a Function Essential for Iron-limited Growth in the Cyanobacterium Anacystis nidulans R2

Summary

We describe the cloning, sequencing and mutagenesis of an Anacystis nidulans R2 gene whose function is required for growth in iron-limited medium. The gene, designated irpA, was cloned from a λgt11 phage expression library: irpA directs the synthesis of a 36 kDa membrane polypeptide which accumulates only in iron-deficient cells. DNA sequence analysis revealed an open reading frame capable of encoding a polypeptide of 38.6 kDa, and hydropathy plots suggested that the hodrophobic amino terminus of the IrpA protein could represent a signal sequence. A site-directed mutant of A. nidulans R2 lacking irpA function was constructed by Tn5 insertion, and the mutant, designated A. nidulans K7, was defective in growth in low-iron medium. Lastly, the DNA sequence of the region immediately upstream from the irpA bears close similarly to the consensus operator sequence conferring iron regulation in Escherichia coli.
