Use of Bovine Alveolar Macrophages and of Their Lysosomes for in Vitro Studies with Mineral Dusts

Abstract

The alveolar macrophage and its lysosomal system play an important role in many physiological and toxicological processes. The exocytosis or intracellular release of lysosomal material probably stand at the beginning or are accompanying a sequence of pathogenetic events leading to cell death and lung injury. For mechanistic studies with cell organelles as well as for repeated studies with genetically homogeneous cells it is of great advantage to work with large numbers of cells (109 or more). In the case of cells from the monocyte-macrophage system such large numbers can often not be obtained. Desai and Richards (1) have used sheep alveolar macrophages recovered by post-mortem lavage for studies with asbestos. In view of the easiness to obtain bovine lungs, which contain still larger numbers of macrophages, we have started to characterize bovine alveolar macrophages (BAM), their cell organelles and their reaction to particles like Latex, TiO2 or quartz taking advantage of the experiences of previous authors (e.g. 2) with these cells.
