Structure of Bacteriorhodopsin and Halorhodopsin in Relation to the Pumping function

Abstract

Bacteriorhodopsin (BR) is a protein present in differentiated patches in the cellular membrane of a highly halophilic microorganism, Halobacterium halobium [1–3]. BR spans the cellular membrane and transports protons from the inside of the cell to its outer milieu upon receiving light energy into its retinylidene chromophore, which is linked to the lysine residue through the protonated Schiff base. The light-dependent proton pumping of BR is driven by two successive processes. The first is the release of proton from the membrane into the outer milieu to which the N-terminus of BR faces. The following is the uptake of proton from the opposite side of the membrane. As a whole, a concentration gradient for proton is established across the membrane. The present article summarizes the studies on the mechanism of energy conversion in BR.
