Abstract
Microtubules are protein fibers that form part of the cytoskeleton of eukaryotic cells. They have a variety of motile and structural functions, for example in mitosis, flagellar movement, or intracellular transport (see Dustin, 1984 and de Brabander & de Mey, 1985 for recent reviews). The following abstract is a brief summary of some results from the literature that are important for understanding current concepts of microtubule structure and assembly. In my contribution I will focus on the application of two biophysical methods, time-resolved X-ray scattering from solutions using synchrotron radiation (see Mandelkow et al., 1980; Bordas et al., 1983) and cryo-electron microscopy of frozen-hydrated molecules (Mandelkow & Mandelkow, 1985).
