Abstract

My group has been especially interested in developing fast reaction techniques that are suitable for making chemical kinetic measurements with receptor-rich membrane vesicles. More recently we have begun to develop techniques for making similar chemical kinetic measurements of receptor function directly on cell surfaces. We are using the information we have already obtained about the mechanism of action of the acetylcholine receptor to calibrate the new methods, and hope that we can then apply the new methods to investigations of many other neuronal receptor proteins, whose study has been hampered by the lack of the equivalent of the electric organ of fish. Most of these receptor proteins exist in very low concentrations and have been characterized so far only by the binding of a specific neurotransmitter. Mechanistic studies have been almost impossible. We are also developing methods to amplify the quantities of such receptor proteins. The Table is an outline of the topics to be considered in this chapter.
