Molecular Mechanism of an Ecotropic MuLV Restriction Gene Akvr-1/FV-4 in California Wild Mice

Abstract

The Akvr-1 restriction gene was discovered as a dominant allele segregating in a feral population of mice (M. domesticus) at a squab farm near Lake Casitas (LC) in Southern California (Gardner et al. 1980). This gene prevented viremia of AKR endogenous retrovirus and virus-mediated lymphoma in LC (Akvr-1RR × AKR) F1 hybrids. This effect was not due to the Fv-1 gene, which prevents AKR viremia and associated lymphoma in lab mice (Pincus et al. 1977), because LC viruses are N-tropic and LC mice appear to be monomorphic for Fv-1N (Gardner 1978). The Akvr-1R gene conferred a strong resistance to in vivo and in vitro infection of various ecotropic murine leukemia viruses (MuLV) (N-tropic, B-tropic or NB tropic) but did not restrict infection with wild mouse amphotropic MuLV strains (Rasheed and Gardner 1983). In these properties, Akvr-1R closely resembled the Fv-4R allele previously identified on chromosome 12 in certain Japanese inbred (FRG strain) and feral mice (M. molossinus) (Odaka et al. 1980; Odaka et al. 1981). Genetic crosses between LC (Akvr-1RR) and FRG (Fv-4RR) mice showed that these resistance loci were allelic (O’Brien et al 1983).
