Early Intestinal Lesions of Graft-vs-Host Reaction and Allograft Rejection in Rodents, Identified by Quantitative Histological Techniques

Abstract

In our investigations of intestinal mucosal lymphocytes, we have used models of T- cell-mediated, delayed-type hypersensitivity (DTH) reactions in the small intestinal mucosa of rodents. Secondary effects in addition to DTH have been reduced by transplanting sterile foetal tissue and by avoiding irradiation in animals with graft-vs- host reaction (GvHR). The evolution of the pathology of the reactions has been examined in various ways, using quantitative techniques whenever possible. The epithelium has been studied by measuring villi, crypts, crypt cell production rate (CCPR) by a stathmokinetic technique, cell migration rate and the number of crypts associated with each villus. Enterocytes have been examined by electron microscopy, assays of brush border enzymes and measurements of labelled amino acid uptake. In the case of lymphoid cells, intraepithelial lymphocytes (IEL) have been counted in all experiments, work on mast cells is described in a separate paper (see A. Ferguson, et al., this volume), and we are currently investigating plasma cells and IgA in these models.
