Zusammenfassung
In einer Übersieht werden verschiedene, einfache Techniken zur epidermalen DNS-Markierung mit radioaktiv markiertem Thymidin miteinander verglichen. Die praktischen Möglichkeiten der in vivo-und in vitro-Methoden werden besprochen. Nachfolgend werden die zellkinetisch bestimmbaren Parameter und schließlich die Ergebnisse der in den Histoautoradiogrammen gewonnenen Daten diskutiert. Die Radioaktivität der verwendeten Isotope schränkt ihren Einsatz bei in vivo-Untersuchungen der menschlichen Haut ein. Im Tiermodell sahen wir eine gute Übereinstimmung zwischen verschiedenen in vivo-Markierungsverfahren und der in vitro-Doppelmarkierungstechnik. Deshalb dürfte diesem in vitro-Verfahren eine wertvolle Rolle bei zytokinetischen Studien auch menschlicher Epidermishyperplasien zukommen. Abschließend haben wir mit Hilfe des in vitro-Doppelmarkierungsverfahrens den Einfluß von UV-Licht auf die epidermale Zellkinetik normaler und psoriatischer Haut untersucht.
Summary
The aim of this article is to review the application to human epidermis of simple DNA-labelling procedures with radioactively labelled thymidine. Practical aspects of in vivo and in vitro labelling methods will be discussed. Subsequently the calculation of kinetic parameters from autoradiographic counting procedures will be considered and finally the relevance of the data so obtained. Especially because of the ethical problems autoradiographic techniques require restrictive application of radioactive substances in human skin in vivo. In animal epidermis a good agreement was seen between several labelling techniques in vivo and double labelling in vitro. Therefore in vitro double labelling methods may well have a useful role to play in kinetic studies on human epidermal hyperplasia. Finally we examined the influence of UV-irradiation on epidermal cell kinetics in normal and psoriatic skin in vitro.
