Zusammenfassung
Zytostatisch wirkende Medikamente und Immunsuppressiva unterdrücken die Vermehrung von Zellen — auch von immunkompetenten — durch Hemmung der DNS-synthetisierenden Enzyme (Glukokortikoide), durch Konkurrenz in der Nukleinsäuresynthese (wie z. B. die Substratkonkurrenz von 6-Mercaptopurin oder Azathioprin mit 6-Aminopurin und 6-Hydroxypurin) und durch Vernetzung der DNS-Stränge (Alkylanzien). Zugleich wird durch diese Substanzen die immunologische Reaktionsfähigkeit, die sog. Immunsurveillance, gestört (Ehrlich, Bernet). Es gibt Hinweise dafür, daß Zytostatika und Immunsuppressiva je nach Art, Dauer und Dosierung karzinogen oder kokarzinogen wirken können. Diese Wirkung kommt wahrscheinlich durch Unterdrückung der Immunreaktionen auf spontan entstehende Tumoren als auch durch onkogene Virusinfektionen zustande, die eine Tumorentstehung fördern können (Promotoren).
