Abstract
The molecular biology of adenoviruses is understood in considerable detail. This relates not only to the primary structure of the viral genome, transcription patterns, and virus/host cell interactions, but also to the mechanism of viral DNA replication. This review will document that adenovirus DNA replication occurs via a novel mechanism for the initiation reaction. In addition, adenovirus replication also represents the only known system that can be initiated efficiently in vitro.
