Nafazatrom: A New Antithrombotic Compound

Abstract

Nafazatrom (BAY G 6575), 3-methyl-l-[2-(2-naphthyloxy)-ethyl]-2-pyra-zolin-5-one, is under development as an antithrombotic drug with a novel mode of action [1]. Its antithrombotic activity was demonstrated in experimentally induced thrombosis, especially in models based on the following freezing technique. The blood vessels of rats, rabbits, and guinea pigs were exposed and dissected free from the surrounding tissue. The jugular vein and the carotid artery of rats or guinea pigs and the femoral arteries of rabbits were used in most of the experiments. The vessels were frozen at −12° C to −15° C over a length of approximately 1 cm in a metal groove for 2–5 min and simultaneously compressed by a weight of 200 g (rat, guinea pig) or 500 g (rabbit). In addition, arteries were clamped with a small silver clip (diameter 0.15–0.20 mm) distally to the lesion. The experimental device is described elsewhere [2].
