Adjuvant Arthritis: The Role of Microbial Cell Wall Products and Other Substances in the Genesis of Joint Disease in Rats

Abstract

Adjuvant arthritis in the rat can be induced by a single injection of a dispersion of certain dried, heat-killed microorganisms such as mycobacteria, Corynebaoteria, streptococci, or their cell wall components such as peptidoglycan and certain peptidoglycan subunits in a suitable oily vehicle [1–4], Some of the bacterial and bacteria-derived substances that have been reported to have arthritogenic activity are listed in Table 1. To be arthritogenic, these substances must be administered in the form of an emulsion in an oily vehicle. Some of the lipid substances that have been found to be suitable for this purpose are summarized in Table 2. Since these oily substances are not arthritogenic by themselves and most of them are biologically inert, they probably serve primarily to modulate (rather than determine de novo) the intrinsic activity of the arthritogenic materials residing in the bacterial cell wall. The mechanism(s) through which the bacterial cell wall materials induce arthritis is not yet clearly understood. There is an impressive body of evidence that the disease arises from an aberrant immune response [5–7] involving primarily the cell-mediated (delayed) hypersensitivity [8, 9], although the humoral immune response may also play a role at an early stage of the disease development [10].
