Zusammenfassung
Es ist inzwischen allgemein akzeptiert, daß auch im menschlichen Plasma ein hö-hermolekulares, cnzymatisch inaktives Renin zirkuliert, das jetzt meist als Prorenin bezeichnet wird (Boyd 1977; Day u. Luetscher 1975; Derkx et al. 1976; Leckie et al. 1977; Rumpf et al. 1978; Skinner et al. 1975). Im normalen Plasma liegt weit über die Hälfte des Gesamtrenins als Prorenin vor, im Durchschnitt etwa 60–85%, so daß nur ein geringer Anteil des zirkulierenden Rcnins tatsächlich enzymatisch wirksam ist (Derkx, et al. 1976; Rumpf et al. 1978). Die Rolle, die das Prorenin - auch inaktives Renin genannt - unter physiologischen und pathophysiologischen Bedingungen spielt, ist unklar. Es gibt jedoch Hinweise auf unterschiedliche Regulationsmechanismcn für die beiden Reninformen (Rumpf etal. 1978). Ungeklärt ist bisher ebenfalls die Frage, ob und - wenn ja - wie das Prorenin in vivo in aktives Renin umgewandelt wird. Die vorliegenden Untersuchungen befassen sich speziell mit dieser Frage. Die Ergebnisse machen eine In-vivo-Aktivierung des Prorenins wahrscheinlich und geben Hinweise auf den der In-vivo-Aktivierung zugrundeliegenden Mechanismus.
