Untersuchungen zum Wirkungsmechanismus von Acetazolamid und SITS auf den Bikarbonattransport im proximalen Tubulus der Rattenniere

Zusammenfassung

Soweit wir heute wissen, erfolgt die Bikarbonatresorption im proximalen Tubulus der Niere durch Sekretion von H+-Ionen in das Tubuluslumen und Rückdiffusion von CO2 aus dem Lumen in die Tubuluszellen. Nach der klassischen Vorstellung hemmen Sufonamide wie Acctazolamid die Bikarbonatresorption, indem sie die Carboanhydratase im Zytoplasma der Nierenzellen blockieren. Dadurch wird die Umsetzung von OH- und CO2zuHC03 - im Zytoplasma verlangsamt, der intrazelluläre pH-Wert steigt an und die H+-Sekretionsrate nimmt ab, weil die H+-Ionen einen größeren Gradienten überwinden müssen.
