G. Wilcox und M.A. Beaven (1976); beschreiben einen einfachen radiochemischen Test für die DBH im Serum. Ying-Hsiueh Wu (1973) beschreiben eine spektroskopische COMT-Bestimmung: Das durch Verlust der Methylgruppe aus S-Adenosyl-methionin (SAM) gebildete S-Adenosylhomocystein (SAH) wird durch ein Enzym (Adenosindesaminase) zu S-Inosylhomocystein (SIH) desaminiert. Die Differenz der Extinktion von SAH und SIH im Ultravioletten soll der Aktivität der COMT proportional sein (A continuous spectrophotometric assay for catechol-O-methyltransferase. Eine fluoreszenzspektroskopische Methode zur Messung der COMT-Aktivität geben O.J. Broch und H.C. Guldberg (1971) an (On the determination of catechol-O-methyltransferase activity in tissue-homogenates. Acta Pharmacol Toxicol 30:266–277). COMT methyliert 3.4-Dihydroxyphenylessigsäure, die nach papierchromatographischer Reinigung und Reaktion mit K-hexacyanoferrat(III) fluorimetrisch bestimmt werden kann.

P.H. Wu und L.E. Dyck (1976) modifizierten die radiochemische MAO-Bestimmung, indem sie überschüssiges radioaktives Substrat an einen flüssigen Ionenaustauscher binden und die desaminierten Reaktionsprodukte direkt in die Szintillations-Flüssigkeit extrahieren. Die Methode soll für verschiedene Amine als Substrat gleichermaßen geeignet sein und sich durch Empfindlichkeit und Schnelligkeit auszeichnen (Microassay for the estimation of monoamine oxidase activity. MAO desaminiert p-Dimethylaminobenzylamin oxidativ zu p-Dimethyl-aminobenzaldehyd, dessen Absorption im Ultravioletten zur quan-titativen Bestimmung ausgenutzt wird (Deitrich RA, Erwin VG (1969) A convenient spectrophotometric assay for monoamine oxidase. Anal Biochem 30:395–402).
Das in der Monoaminoxidase-Reaktion entstandene Wasserstoffperoxid bildet in Gegenwart von Peroxidase aus Leuco-2′,7′-dichlor-fluoreszein das farbige 2′,7′-Dichlorfluoreszein, dessen Absorption bei 502 nm der MAO-Aktivität proportional sein soll (Köchli H, von Wartburg JP (1978) A sensitive photometric assay for monoamine oxidase. Anal Biochem 84:127–135).
Dasselbe Prinzip benutzt K.F. Tipton (1969) für eine MAO-Bestimmung: Homovanillinsäure wird durch H2O2 und Peroxidase zu einem fluoreszierenden Produkt umgewandelt. Der Vorteil dieser Methode ist die höhere Empfindlichkeit aufgrund der fluoreszenzspektro-skopischen Messung (A sensitive fluorometric assay for monoamine oxidase. Anal Biochem 28:318–325).

In Analogie zur TOH-Bestimmung setzen W. Lovenberg, R.E. Bensinger, R.L. Jackson und J.W. Daly (1971) 5–3H-DL-Tryptophan als Substrat der TrpOH ein. Das Reaktionsprodukt 4–3H-5-Hydroxytryptophan tauscht das Tritium mit Wasser aus, so daß die Menge des gebildeten 3H-H2O der TrpOH-Aktivität direkt proportional ist (Rapid analysis of tryptophan hydroxylase in rat tissue using 5–3H-tryptophan. Anal Biochem 43:269–274).
J.S. Kizer, J.A. Zivin, J.M. Saavedra und M.J. Brownstein (1975) beschreiben einen komplizierten Test zur Bestimmung der TrpOH-Aktivität, der eine vielstufige enzymatische Umsetzung des L-Tryptophans umfaßt. Die beiden letzten Reaktionen sind Bestandteil der radioenzymatischen Serotonin-Bestimmung von Saavedra et al. (s. dort) (A sensitive microassay for tryptophan hydroxylase in brain. Eine neuere Arbeit beschreibt eine Serotonin-Bestimmung, die auf demselben Prinzip wie die von Saavedra et al. angegebene beruht. Die N-Acetylierung des Serotonins wird hier chemisch mit Acetan-hydrid durchgeführt. Die Autoren geben als Vorteil ihrer Methode an: Größere Empfindlichkeit durch höheren Acetylierungsgrad und niedrigere Blindwerte durch fehlende Decarboxylase-Aktivität (in teilgereinigter Arylamin-N-Acetyltransferase enthalten) (Hammel I, Naot Y, Ben-David E, Ginsburg H (1978) A simplified microassay for serotonin: modification of the enzymatic isotopic assay. Anal Biochem 90:840–843).
Zahlreiche fluorimetrische Bestimmungsmethoden für Serotonin sind beschrieben worden, von denen hier zwei angegeben werden sollen. Snyder SH, Axelrod J, Zweig M (1965) A sensitive and specific fluorescence assay for tissue serotonin. Biochem Pharmacol 14: 831–835. Nach Extraktion des Serotonins mit organischen Lösungsmitteln läßt man es mit Ninhydrin zu einem Produkt reagieren, dessen intensive Fluoreszenz quantitativ ausgewertet wird.
C. Atack und M. Lindqvist (1973) nutzen die Reaktion mit o-Phthaldialdehyd zur fluorimetrischen Serotonin-Bestimmung aus (Conjoint native and orthophthaldialdehyde-condensate assays for the fluorimetric determination of 5-hydroxyindoles in brain. Naunyn-Schmiedebergs Arch Pharmacol 279:167–284).

In mehreren gekoppelten enzymatischen Reaktionen wird cAMP zu ATP umgewandelt, das in Gegenwart von Sauerstoff mit einer Luciferin-Luciferase-Präparation (aus amerik. Leuchtkäfer) unter Lichtemission reagiert. Eine quantitative Auswertung dieses emittierten Lichtes ist u.a. im Flüssigkeits-Szintillationsspektrometer möglich (The luminescence assay of cyclic AMP. Anal Chem 43:1701–1703).
P.S. Schönhöfer et al. (1972) setzen 32P-cAMP als Substrat der PDE ein. Eine Phosphatase dephosphoryliert das gebildete 32P-AMP zu Adenosin. Das freiwerdende 32P-Phosphat kann nach Komplexierung mit Molybdat extrahiert werden (Cyclic 3′,5′-AMP phosphodiesterase in isolated fat cells. Pharmacology 7:65–77).
Eine einfache und schnelle Methode zur PDE-Bestimmung wird von D.R. Zusman (1978) angegeben. Das aus 3H-cAMP entstandene 3H-AMP wird auf Celluloseplatten durch Waschen mit LiCl-Lösung vom Ausgangsprodukt getrennt (A rapid batch assay for cyclic AMP phosphodiesterase. Anal Biochem 84:551–558).

Nach Adsorption des 32P-Histonphosphats an Cellulosephosphat-Papier werden 32P-ATP und wasserlösliche Metabolite ausgewaschen und das phosphorylierte Histon im Szintillations-Spektrometer gezählt (Rapid protein kinase assay using phosphocellulose paper absorption.
