Abstract

This Chapter is not intended to be a comprehensive literature survey, but rather an introduction to techniques that are now available for preparing nucleic acid samples for electron microscopy. In particular, an effort has been made to emphasize a quantitative approach to the electron microscopy of nucleic acids and to give examples of quantitative applications of the techniques discussed. We have often used examples from our own research to demonstrate ways in which quantitative electron microscopy can be done; however, this does not imply that these are the only examples available. Younghusband and Inman (1974) published a review of the electron microscopy of DNA that contains extensive literature references, and Chow et al. (1975) prepared a laboratory manual for the electron microscopy of nucleic acids containing extensive information about techniques, instruments, and supplies.
