Zusammenfassung
Glukagon, der hyperglykämisierende glykogenolytische Faktor des Pankreas, wurde fast zur selben Zeit wie das Insulin entdeckt, nämlich vor ungefähr 5 Jahrzehnten. Kimball und Murlin (1923) trennten als erste eine hyperglykämisierende Substanz aus Insulinpräparaten ab, die sie Glukagon nannten. Neben vielen anderen haben de Duve (1953) und Bergen und van Itallie (1960) die Geschichte der Glukagonforschung ausführlich beschrieben. Während der 20 Jahre, die seiner Entdeckung folgten, kamen nur wenig neue Erkenntnisse über das Glukagon hinzu. Manche Forscher, wie Bürger u. Brandt (1935) glaubten an seine Existenz und versuchten, das Glukagon in reiner Form darzustellen. Demgegenüber schrieben andere die hyperglykämisierenden und glykogenolytischen Wirkungen der Pankreasextrakte und einiger Insulinpräparate dem Insulin selbst oder der Anwesenheit von heterogenen Verunreinigungen zu. Erst in den fünfziger Jahren trat die Arbeit über das Glukagon in eine neue Phase, als Sutherland u. de Duve (1948) und Sutherland et al. (1949) den hyperglykämisierenden Faktor aus dem Pankreas extrahierten und begannen, seine Eigenschaften zu untersuchen. Seitdem ist eine Fülle von Informationen über die verschiedenen metabolischen Wirkungen und den Wirkungsmechanismus dieses Peptids gesammelt worden. Trotzdem war es lange Zeit schwierig, die Stellung des Glukagon als Hormon und seine physiologische Rolle zu definieren, insbesondere beim Menschen. Dies hing hauptsächlich mit der Tatsache zusammen, daß bis vor kurzem kein definierter pathologischer Zustand einem Überschuß oder einem Defekt der Glukagonsekretion zugeordnet werden konnte.
