Zusammenfassung
Es werden die Reaktionen des arzneimittelabbauenden Enzymsystems der Rattenleber auf wiederholte Halothan- und Ēthrane-Gaben in narkotischen und subnarkotischen Konzentrationen vergleichend untersucht. Leberfunktionsteste, Hexobarbitalschlafzeit, N -Demethylierung von Äthylmorphin, NADPH-Cytochrom-c-Reduktaseaktivität und der Gehalt an Cytochrom P450 der Leber wurden am 1., 3. und 7. Tag nach der letzten Gabe ermittelt.
Folgende Ergebnisse wurden gewonnen:

Wiederholte Halothan- und Ēthrane-Inhalationen in narkotischen Konzentrationen beeinflussen das mikrosomale Enzymsystem der Rattenleber im Sinne einer Enzyminduktion. Die Veränderungen des Leberfeuchtgewichtes, der Hexobarbitalschlafzeit und der NADPH-Cytochrom-c-Reduktaseaktivität sind nach Ēthrane geringer als nach Halothan, während der Gehalt an Cytochrom P450 durch beide Narkotika nicht vermehrt wurde.

Subnarkotische Konzentrationen von 0,3 Vol.-% Halothan, täglich 2 Std über vier Wochen, erhöhen das relative Leberfeuchtgewicht, verkürzen die Hexobarbitalschlafzeit und steigern die N-Demethylierung von Äthylmorphin sowie die NADPH-Cytochrom-c-Reduktaseaktivität signifikant. Bei Zufuhr von Ēthrane in subnarkotischen Konzentrationen von 0,6 und 0,8 Vol.-% ist nur bei der höheren Konzentration eine geringe Verkürzung der Hexobarbitalschlafzeit und eine mäßige Beschleunigung der N-Demethylierung von Äthlymorphin zu beobachten.

Nach Vorbehandlung mit 0,5 Vol.-% Halothan bzw. 0,6 Vol.-% Ēthrane nimmt die Ausscheidungs- und Bindungskapazität der Leber für BSP nach Halothan stärker zu als nach Ēthrane. Auch wenn schädliche Wirkungen dieser Adaptationsphänomene auf den Organismus zur Zeit allgemein negiert werden, kann eine Persistenz gesteigerter Enzymaktivitäten über einen längeren Zeitraum die Elimination und Wirkungsdauer anderer Pharmaka beeinflussen.
Summary
The reactions of the enzyme systems in the rat liver responsible for the breakdown of pharmaceuticals were studied under conditions where Halothane and Ēthrane were given repeatedly in narcotic and subnarcotic doses. Liver function tests were carried out and hexobarbital sleeping time, N-demethylation of ethyl morphine, and the liver NADPH-cytochrome c reductase activity and cytochrome P450 were determined on days 1, 3 and 7. The following results were obtained:

Repeated inhalation of Halothane and Ēthrane at narcotic concentrations caused enzyme induction in the microsomal enzyme system of rat liver. Changes in wet weight of liver, hexobarbital sleeping time, and the NADPH-cytochrome c reductase activity were less after Ēthrane than Halothane, while the cytochrome P450 values were not influenced by either compound.

Subnarcotic doses of Halothane at concentrations of 0.3 vol.-% for two hours daily over aperiod of four weeks significantly increase the relative wet weight of liver, shorten hexobarbital sleeping time and N-demethylation of ethyl morphine, as weH as NADPH-cytochrome c reductase activity. When Ēthrane is given at the subnarcotic concentrations of 0.6 and 0.8 vol.-%, at the higher dose only a slight reduction is observed in hexobarbital sleeping time and in the increase of N-demethylation of ethyl morphine.

After pretreatment with 0.5 vol.-% Halothane or 0.6 vol.-% Ēthrane, the BSP elimination and binding capacities of the liver are more markedly increased with Halothane than Ēthrane. Assuming the harmful effects of these adaptation phenomena on the organism at this time were totally eliminated, the persistence of increased enzyme activity over a longer period of time can affect the elimination and duration of activity of other pharmaceuticals.
