Abstract

The molecular processes associated with DNA replication1 and cell division2, the mechanisms involved in the control of cell proliferation, and the kinetic analysis of cell proliferation in simple and complex cell systems are topical problems and the subject of current research in cell biology. Although the principal mechanisms that regulate cell proliferation in mammalian cell systems in vivo are still largely unclarified, appreciable advances have been made in the study of the proliferative characteristics of tissues in terms of cell population kinetics. Yet, despite the many experimental and theoretical contributions to this area of research, the information presently available—particularly in the case of neoplastic tissues—is still incoherent and does not allow a definitive picture to be drawn. This review sums up the present state of knowledge about the proliferative characteristics of malignant cell systems, with particular emphasis on solid neoplasms. Various aspects of the cell (replicative) cycle, together with the methods used for the analysis of cell proliferation kinetics in normal and malignant cell systems, have been described in a number of reviews and conference proceedings3–22. No attempt is therefore made to present a complete compilation of data from the literature; the aim is rather to indicate where conceptual and methodological difficulties still exist, or where further experimental evidence is required for clarification.
