3’-Deoxyadenosine and Other Polynucleotide Chain Terminators

Abstract

At present only one principle for enzyme reactions responsible for synthesis of polynucleotide chains is known. It consists of extension of chains in the 5’→3’ direction by a nucleophilic attack of the 3’-0H group of the 3’-terminal nucleotidyl group on the phosphorus atom of the α-phosphate group of a nucleoside di- or triphosphate, resulting in extension of the chain by a further nucleoside monophosphate. The incorporation into a polymer by this type of reaction of a nucleoside monophosphate lacking the free 3’-OH group in the ribose configuration would prevent further chain elongation since no further 3’, 5’-phosphodiester linkage can be formed. This expectation has been experimentally confirmed. Any nucleoside or nucleotide containing this type of sugar moiety could, therefore, be regarded as a potential polynucleotide chain growth terminator, and will be the topic of the present chapter.
