Cytotoxic Inhibitors of Protein Synthesis

Abstract

Many antineoplastic agents exert profound inhibitory effects on protein synthesis. With few exceptions, such drugs are too toxic to be used for therapeutic purposes, but many have been employed as experimental tools in cell biology. Several reviews and symposia have been devoted to this topic (Goldberg, 1965; Gottlieb and Shaw, 1967; Bucher and Sies, 1969; Pestka, 1971; Munoz et al., 1972; Grollman and Huang, 1973), and this chapter will attempt to summarize current knowledge of the biochemical pharmacology of these drugs.
