Abstract
Tests predictive of cytotoxicity are of three general types; those that select agents for antitumor cytotoxicity, those that predict host cytotoxicity, and those applied to predict the toxicity/benefit ratio of a drug of generally accepted potency in a particular patient with a specific tumor. The first type is fundamental to the other two, since the agent involved must be able to inhibit cell growth in order to deserve any consideration, and agents that show such activity will then be studied in more detail for their effects upon the tumor-bearing host. The latter two types of tests are basic, therefore, respectively, to screening of potential new drugs in subhuman systems and to initial human trials. Screening for tumor cytotoxicity has been discussed in Chapter 2 of this volume, and many of the considerations in regard to host toxicity of known effective agents have been discussed in Chapter 18. The present chapter will, therefore, deal primarily with tests predictive of antitumor toxicity of a specific drug in a specific patient.
