Abstract

One of the oldest and most commonly used procedures in biological research to elucidate the physiological significance of an organ is its destruction or functional elimination. This principle has found wide application in the investigation of the physiological role of endocrine glands, in the characterization of the coordinative and regulatory function of different centres and tracts of the central nervous system and also in the study of the peripheral nervous system. Particularly for the sympathetic nervous system this procedure has proved to be most important in the elucidation of its general homeostatic function (Cannon et al., 1929; Sawyer et al., 1933; Cannon and Rosenblueth, 1937) and in the more detailed investigations concerning the sites of synthesis, storage and enzymatic degradation of the adrenergic transmitter (Von Euler and Purkhold, 1951; Goodall, 1951; BURN et al., 1954; CROUT and Cooper, 1962; Anden et al., 1964; Waltman and Sears, 1964; Hertting, 1965; Klingman, 1965; Potter et al., 1965; Iversen et al., 1966, 1968; Sedvall and Kopin, 1967; Mueller et al., 1969; Nagatsu et al., 1969). Moreover, denervation experiments have been of crucial importance in detecting that uptake into the adrenergic nerve terminals is generally the main mechanism by which exogenously administered or neurally liberated noradrenaline is inactivated (Hertting et al., 1961a; Hertting and Schiefthaler, 1964; Hertting, 1965; Potter et al., 1965) and that the activity of the adrenergic neurons beyond their actual effect have also “trophic” actions on the effector organs (BURN, 1952; Trendelenburg and Weiner, 1962; Fleming, 1963; Trendelenburg, 1963a, b).
