Deficiencies of the MHC Class I Antigen Processing and Presentation Machinery in Tumor Cells

Abstract

The major histocompatibility complex (MHC) class I genes, which are composed of a polymorphic heavy chain noncovalently linked with ß2-microglobulin (ß2-m; Bjorkman et al.1987), play a key role in numerous immunological processes involving the presentation of “foreign” antigens for recognition by the CD8-positive cytotoxic T lymphocytes (CTLs; Zinkernagel and Doherty 1979). This suggests that (1) cytotoxic T cells can monitor genetic changes and that (2) the MHC class I system provides an enormous diversity in potential antigenic peptide presentation including recognition of tumor cells by autologous CTL in man and animals (Boon et al. 1994).
